This study is a Phase III, randomised, open label, multi-centre study assessing the efficacy and safety of MEDI4736 versus Standard of Care in NSCLC patients with PD-L1 positive tumours and the combination of MEDI4736 plus tremelimumab (MEDI4736+treme) versus Standard of Care in NSCLC patients with PD-L1-negative tumours in the treatment of male and female patients with locally advanced or metastatic NSCLC (Stage IIIB-IV), who have received at least 2 prior systemic treatment regimens including 1 platinum-based chemotherapy regimen for NSCLC. Patients with known EGFR (Epidermal growth factor receptor) tyrosine kinase (TK) activating mutations and anaplastic lymphoma kinase (ALK) rearrangements are not eligible for the study (prospective testing is not planned within this study). The Standard of Care options are: an EGFR tyrosine kinase inhibitor (erlotinib [TARCEVA®]), gemcitabine or vinorelbine (NAVELBINE®). Understanding the Relationship Between Benign Breast Tissue, Benign Breast Disease, and Breast Cancer Development Jacksonville, Fla. The purpose of this study is to culture human mammary cells to identify cellular characteristics associated with lobular involution status. A Study of ONO-7475 in Patients With Acute Leukemias Jacksonville, Fla. The purpose of this study is to assess the safety and tolerability of ONO-7475 monotherapy in patients with relapsed or refractory acute myeloid leukemia or relapsed or refractory myelodysplastic syndromes, and to assess the safety, tolerability, and preliminary effectiveness of the combination of ONO-7475 and venetoclax in patients with relapsed or refractory acute myeloid leukemia. Pre-op Pembro + Radiation Therapy in Breast Cancer (P-RAD) Rochester, Minn. The purpose of this trilal is to study a combination of neoadjuvant radiotherapy (RT), immunotherapy (pembrolizumab) and chemotherapy for lymph node-positive, triple negative (TN) or hormone receptor positive/HER2-negative breast cancer. A Registry to Collect Patient and Family DNA and Information for Anyone who has had Multiplex Testing and Gene Mutation Results Rochester, Minn. The purpose of this study is to collect patient and family DNA and disease data to learn more about how changes in certain genes may be linked to cancer. Some people with cancer may have aquired their disease because they inherited an abnormal (mutated) gene. The researchers of this study want to better understand the risks that are linked to genetic changes in these less well-studied genes. By understanding these risks, it is believed that doctors will be able to give better advice to families with mutations in these genes. A Study to Evaluate Exemestane in Post-Menopausal Women with Non-Small Cell Lung Cancer (NSCLC) Albert Lea, Minn., Mankato, Minn. This study is being conducted to see if adding Exemestane to the immune checkpoint blockade can slow disease progression in post-menopausal women with non-small cell lung cancer. Reduced Craniospinal Radiation Therapy and Chemotherapy in Treating Younger Patients With Newly Diagnosed WNT-Driven Medulloblastoma Rochester, Minn. This phase II trial studies how well reduced doses of radiation therapy to the brain and spine (craniospinal) and chemotherapy work in treating patients with newly diagnosed type of brain tumor called WNT)/Wingless (WNT)-driven medulloblastoma. Recent studies using chemotherapy and radiation therapy have been shown to be effective in treating patients with WNT-driven medulloblastoma. However, there is a concern about the late side effects of treatment, such as learning difficulties, lower amounts of hormones, or other problems in performing daily activities. Radiotherapy uses high-energy radiation from x-rays to kill cancer cells and shrink tumors. Drugs used in chemotherapy, such as cisplatin, vincristine sulfate, cyclophosphamide and lomustine, work in different ways to stop the growth of tumor cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Giving reduced craniospinal radiation therapy and chemotherapy may kill tumor cells and may also reduce the late side effects of treatment. A Study to Evaluate Immunotherapy and Ovarian Function Among Pre-menopausal Melanoma Survivors Rochester, Minn. The purposes of this study are to analyze the ovarian function of female premenopausal melanoma survivors who have undergone immunotherapy, and compare with data from age-matched controls, prospectively investigate ovarian function in premenopausal women with melanoma undergoing immunotherapy, and to identify the impact of ovarian function change on the frequency and function of CD8+ T cells during immunotherapy. Tissue Collection for Patients with Cancers other than Hepatobilairy Cancers with Metastasis to the Liver Rochester, Minn. The goal of this study is to develop a resource (bank) of biospecimens (blood, tissue) from patients having cancers that metastasis to the liver. The purpose of the resource bank is to facilitate studies on the pathogenesis of cancers metastasizing to the liver and also in order to assess the ability of new tests to detect early cancer in the cohort of patients undergoing routine surveillance for cancer. The biospecimens will include cultures of tumor cells and tumor-associated non-malignant cells, as well as xenografts established from freshly isolated metastatic cancers in immunodeficient mice. We plan to administer a risk factor questionnaire to facilitate identification of risk factors for development of metastatic liver cancer. The study will also determine survival of patients from time of diagnosis. Exploring the Role of B-cell Activating Factor Receptor (BAFFR)-based Chimeric Antigen Receptor T-cell (CAR T) in BAFFR-expressing B-cell Hematologic Malignancies and Autoimmune Rheumatologic Disorders Jacksonville, Fla. The purposes of this study are to explore the therapeutic efficacy of BAFFR-CAR T cells in BAFFR-expressing B-cell hematologic malignancies including large B-cell, mantle cell and follicular lymphoma, chronic lymphocytic leukemia (CLL) and B-cell acute lymphoblastic leukemia (B-cell ALL) using primary tumor and/or patient derived xenograft models, and to explore the therapeutic efficacy of BAFFR-CAR T cells in autoimmune rheumatologic diseases including systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), and anti-neutrophil cytoplasmic antibody (ANCA) associated vasculitis using primary samples and/or patient derived xenograft models.
