Research Fellowship - My project investigated the role of VAV, a guanine nucleotide exchanged factor, as a signal transduction molecule involved in the phagocytosis of yeast. Phagocytosis entails the recognition and engulfment of microbes by numerous cell surface receptors. Particle recognition is accompanied by polymerization of the actin cytoskeleton. VAV regulates Rho GTPases, which in turn regulate cytoskeleton rearrangement. My experiments involved phagocytic assays on wild type and VAV-knockout murine macrophages, immunoblot analysis and immunostaining for total and phosphorylated VAV molecules, and creating a model of systemic candidiasis infection to observe the effect of VAV on the innate immune response. In performing these experiments, I was able to show that VAV is essential to the phagocytosis of yeast particles both in vitro and in vivo.Rhode Island Hospital
